Do the three levels of mastery build on each other, or does only the highest level of primer you've read count? Would someone who read a level 3 taming primer have the same abilities as someone who read 1, 2, and 3? Do you have to read them in order?

The higher primers include everything from the lower ones.

You don't need to use 1, 2 and 3, if you already have a 3. Just use the 3 and you have it all.

Is there a way to check which level of mastery you've read?

click on the mastery book and choose "change mastery" - it will list your masteries and their levels
