Can I case Protection on a MOB

I'd like to cast protection on, say, DreadHorn to lower his physical resist further, and give my double strikes more oomph.

Will this work?

Not unless you can persuade Dreadhorn to accept a party invite, then Arch Protection would work, the only things you can do to lower it's Physical resist are Onslaught, Discord, Lesser/Hiryu. If you have a reliable way to cure it's Poison then the Lesser/Hiryu would work well as you could stand on the same tile to keep aggro.
