They'll work as long as something isn't in the way.
Blocking items could be anything ranging from a house placed slightly different from the one you originally marked at, to people banksitting (a big issue for runes taken to Atlantic), to EM-placed items.
But, barring those, you'll find most of your old runes will work on the shards you take them to. As to the safety of your arrival, that's not guaranteed.
